Algal toxin profiles in Nigerian coastal waters (Gulf of Guinea) using passive sampling and liquid chromatography coupled to mass spectrometry ArchiMer
Zendong, Suzie Zita; Kadiri, Medina; Herrenknecht, Christine; Nezan, Elisabeth; Mazzeo, Antonia; Hess, Philipp.
Algal toxins may accumulate in fish and shellfish and thus cause poisoning in consumers of seafood. Such toxins and the algae producing them are regularly surveyed in many countries, including Europe, North America, Japan and others. However, very little is known regards the occurrence of such algae and their toxins in most African countries. This paper reports on a survey of phytoplankton and algal toxins in Nigerian coastal waters. Seawater samples were obtained from four sites for phytoplankton identification, on three occasions between the middle of October 2014 and the end of February 2015 (Bar Beach and Lekki in Lagos State, Port Harcourt in Rivers State and Uyo in Akwa Ibom State). The phytoplankton community was generally dominated by diatoms and...

Bardouil, Michele; Masselin, Pierre; Bohec, Madeleine.
Laboratory assays have been performed, using mussels contamined with Dinophysis, in order to assess potential depurations kinetics. Mussels samples from Seine bay, Douarnenez bay and Vilaine bay have been tested . From these assays, which are to be performed in 1989; two main results are stressed : 1) when the toxicity level is near 0,5 MU/g of hepatopancreas*, depuration is undedectable during twelve days, 2) when the toxicity level exceeds 1 MU/g,depuration is important during the first eight days. *(quarantine level used by Japanese authorities) .

Combined effects of temperature, irradiance and pH on Teleaulax amphioxeia (Cryptophyceae) physiology and feeding ratio for its predator Mesodinium rubrum (Ciliophora) ArchiMer
Gaillard, Sylvain; Charrier, Aurelie; Malo, Florent; Carpentier, Liliane; Bougaran, Gael; Hégaret, Helene; Réveillon, Damien; Hess, Philipp; Séchet, Veronique.
The cryptophyte Teleaulax amphioxeia is a source of plastids for the ciliate Mesodinium rubrum and both organisms are members of the trophic chain of several species of Dinophysis. It is important to better understand the ecology of organisms at the first trophic levels before assessing the impact of principal factors of global change on Dinophysis spp. Therefore, combined effects of temperature, irradiance and pH on growth rate, photosynthetic activity and pigment content of a temperate strain of T. amphioxeia were studied using a full factorial design (central composite design 23*) in 17 individually controlled bioreactors. The derived model predicted an optimal growth rate of T. amphioxeia at a light intensity of 400 µmol photons · m‐2 · s‐1, more...

Amzil, Zouher; Mathias, Amaury.
Ifremer monitoring network for phytoplankton and phycotoxins (REPHY) is in charge of shellfish safety control and carries out shellfish toxicity test on mice as soon as Dinophysis cells (okadaic acid "OA" producer) are observed in sea water. However, this test can neither identify or quantify the diarrhetic toxins OA and its analogues (DTX-1, DTX-3). Only a chemical analysis is able to detect them. At first, the shellfish extraction procedure used for mouse-tests was optimised in order to adjust it for OA/DTX-1 chemical analysis. Due to the Iack of standard, DTX-3 quantification is clone indirectly after a hydrolysis stage releasing OA. Chemical analysis of shellfish samples collected in 2001 from three locations of "Long-line Mussel Farming" of southern...

Gaillard, Sylvain.
Harmful algal blooms of Dinophysis are responsible for diarrheic shellfish poisoning in human consumers of contaminated mollusks after bioaccumulation of their toxins. Dinophysis is a mixotrophic organism, which sequesters chloroplasts from a unique prey Mesodinium rubrum to perform its own photosynthesis, itself mixotrophic and feeds a cryptophyte, Teleaulax amphioxeia. The relationship between these three organisms is fundamental to understand the distribution and bloom dynamics of Dinophysis. In a first study, the effects of temperature, irradiance and pH on physiological aspects of T. amphioxeia were determined using a full factorial design before assessing the effect of contrasting physiological states after acclimation to two light intensities of T....

Explaining dinophysis cf. acuminata abundance in Antifer (Normandy, France) using dynamic linear regression ArchiMer
Soudant, Dominique; Beliaeff, Benoit; Thomas, G.
Classical regression analysis can be used to model time series. However, the assumption that model parameters are constant over time is not necessarily adapted to the data. In phytoplankton ecology, the relevance of time-varying parameter values has been shown using a dynamic linear regression model (DLRM). DLRMs, belonging to the class of Bayesian dynamic models, assume the existence of a non-observable time series of model parameters, which are estimated on-line, i.e. after each observation. The aim of this paper was to show how DLRM results could be used to explain variation of a time series of phytoplankton abundance. We applied DLRM to daily concentrations of Dinophysis cf. acuminata, determined in Antifer harbour (French coast of the English...

First report on amnesic and diarrhetic toxins detection in French scallops during 2004-05 monitoring surveys ArchiMer
Amzil, Zouher; Royer, Florence; Sibat, Manoella; Fiant, Liliane; Gelin, Muriel; Le Gal, Dominique; Francoise, Sylvaine.
In the context of the French phytoplankton and phycotoxins monitoring network (REPHY), shellfish scallops were harvested systematically during the authorised fishing season for the purpose of investigating paralytic toxins (PSP-mouse test), amnesic toxins (ASPchemical analysis), and lipophilic toxins (DSP-mouse test). For all shellfish samples that tested positive in the DSP mouse bioassay used for lipophilic toxins detection, liquid chromatography coupled with mass spectrometry (LC-MS/MS) was used to search for the following lipophilic toxins: okadaic acid, dinophysistoxins, pectenotoxins, azaspiracids, yessotoxins, spirolides, and gymnodimine. In order to investigate the presence of okadaic acid esters (DTX3), alkaline hydrolysis was performed on all...

Lassus, Patrick; Truquet, Philippe; Le Dean, Loic; Bardouil, Michele.
During the oceanographic cruise of June 1988, phytoplankton and correlated hydrological parameters were studied in the coastal area between Camaret and Ile d'Oléron. 62 stations were sampled in the 0-30 m layer within the 50 m depth limited coastal area. Observations focused on two toxic dinoflagellate species: Dinophysis spp and Gyrodinium aureolum. Dinophysis spp is more abundant in the Loire/Vilaine area, which is.characterized by high surface Nitrate levels, high 30 m depth Phosphate levels and Chlorophyll a amounts higher than elsewhere during the survey. This particuliar pattern can be related to the Nutrient outputs of Loire and Vilaine Rivers. Max. Dinophysis concentrations were observed in Vilaine bay (very high Phosphate levels) and in Douarnenez...

Innovative rapid solid-phase immunobead assay for the detection of okadaic acid and related DSP toxins in shellfish ArchiMer
Park, D; Fremy, J; Marcaillou, L; Gamboa, P; Gleizes, E; Masselin, P; Goldsmith, C.
The solid-phase immunobead assay (Ciguatect super(TM)) has been optimized to detect toxins associated with diarrheic shellfish poisoning (DSP). The Ciguatect super(TM) test kit utilizes a membrane to bind the toxins and a monoclonal antibody-latex bead solution as the determination step. The presence or absence of okadaic acid (OA) and related toxins is determined by binding the toxins to a membrane attached to a plastic strip and exposing the toxins ladened membrane to a monoclonal antibody-colored latex bead complex which has a high specificity for OA and related polyether compounds. The intensity of the color on the membrane denotes the presence of the toxins. A rapid extraction method (REM super(TM)) has been developed to allow for confirmation of...

Lassus, Patrick; Bardouil, Michele.
From seawater samples collected between 1984 and 1989 in 12 French coastal areas, a morphometric study have been realised on Dinophysis genera restricted to "acuminata" like species. Simple morphological criterias are given as suitable for discrimination of four dominant species occurring during toxic episodes in shellfish farms (Diarrheic Shellfish Poisoning): Dinophysis c-closed-integral. sacculus, D. c-closed-integral. acuminata, Dinophysis sp. and D. c-closed-integral. norvegica. This last species cannot be included in the "acuminata complex" and produces temporary blooms along southern Brittany coasts. Pending a more precise diagnosis it is recommended to clarify the relationship between toxin composition and each species previously described.

Lassus, Patrick; Maggi, Pierre; Proniewski, F.; Truquet, Philippe; Bardouil, Michele.
Since 1987 Antifer oil tanker terminal (Normandy, France) was weIl known, as a preferential area for summer highest blooms of Dinophysis cf acuminata, a toxic dinoflagellate. Three oceanographic cruises were made in August 1989, before, in the course of and after the seasonal max. of this species. Results obtained from these cruises (hydrological parameters and phytoplankton) as weIl as from a daily survey at a reference station in Antifer harbour, corrobora te the prevailing role of coastal up flowing of Seine estuarine waters, pushed to the coast by a dominant S - SW wind, on intensity and duration of Dinophysis bloom. Such observations are in agreement with those of Summer 1987 at reference station.

Belin, Catherine; Raffin, Bernard.
This document presents the results obtained through the French Phytoplankton and Phycotoxins Monitoring Network (REPHY), concerning the presence of toxic and harmful phytoplankton species along the French coast, from 1984 to 1995. The network REPHY is briefly described. Then the different species, and their associated events, are reviewed : first, species toxic to shellfish consumers, e.g. Dinophysis spp. and Alexandrium minutum, which respectively produce diarrheic and paralytic toxins ; then, species which are toxic or harmful for marine fauna (fish, shellfish). The monitoring results are preceded by the main knowledge on taxonomy, distribution, biology, ecology and toxicity of the concerned species. A final synthesis illustrates the distribution of...

Light and dark carbon uptake by Dinophysis species in comparison to other photosynthetic and heterotrophic dinoflagellates ArchiMer
Graneli, Edna; Anderson, Donald M; Carlsson, Per; Maestrini, Serge Y.
The marine dinoflagellate genus Dinophysis includes species that are the causative agents of diarrhetic shellfish poisoning (DSP). Recent findings indicate that some Dinophysis species are mixotrophic, i.e. capable of both autotrophic and heterotrophic nutrition. We investigated inorganic (and organic) carbon uptake by several species of Dinophysis in the Light and dark using the 'single-cell C-14 method', and compared uptake rates with those of photosynthetic Ceratium species and heterotrophic dinoflagellates in the genus Protoperidinium. Experiments were conducted with water from the Gullmar Fjord and from the Koster Strait (Swedish west coast). Nutrient-enriched phytoplankton from surface water samples were concentrated (20 to 70 mu m) and incubated at...
